Here is a summary of new ideas for the ‘juvenile resistance to stress’ experiment following a discussion with Steven and Brent on Wed. Aug. 7:

I. chronic pH stress (2 months)

  • amb H2O w/ elev. hist.
  • amb H2O w/ amb. hist.
  • var low pH H2O with elev. hist.
  • var low pH H2O with amb. hist.
  • something like this:

II. acute stress (6 hrs, 24 hours, 48 hours?)

  • freeze samples for metabolite(omics?) and epigenetic analysis
  • respirometry?
  • Possible acute stressors:
    • temp
    • salinity
    • temp + salinity
    • lagoon water
    • desiccation

Plan for acute stress:

  1. Test one stress at a time:

  2. Test multiple stressors simultaneously:

    • I’m not sure how realistic respirometry would be for this

III. Measurements to make outside of stress trials

  1. pre-chronic stress baseline measurement
    • freeze samples for metabolite(omics?) and epigenetic analysis
    • respirometry?
  2. pre-acute stress measurement after 1 week chronic pH stress?
    • freeze samples
    • respirometry?

I also went through all the samples taken fall 2018-Spring 2019 and came up with these schematics to better understand the samples we have for possible analyses:
